L-Theanine Benefits: What the Human Trials Actually Found

TL;DR

L-theanine has decent evidence for dampening the physical stress response and modest evidence for sleep quality. The focus claim is shakier than it looks, because most of the attention research tested L-theanine combined with caffeine rather than on its own. Trials used 200 mg for stress and sleep, and 200 to 400 mg in the reviews. Taken alone at higher doses, it has slowed reaction time in one trial and failed to beat placebo for clinical anxiety in another.

What L-theanine is

L-theanine is an amino acid found in tea leaves. It isn't used to build proteins, which is why you'll see it described as a non-protein amino acid, and it's the compound usually credited for the difference between how tea and coffee feel.

It reaches the brain and changes measurable brain activity. In a resting EEG study, 50 mg produced a larger rise in alpha activity than placebo in people sitting quietly with their eyes closed, an effect the authors described as relaxing the mind without inducing drowsiness (Nobre 2008).

The doses the trials used

Every claim below belongs to a dose, so it's worth setting them out first.

Stress and sleep trials in adults have mostly used 200 mg daily (Hidese 2019). A systematic review of nine randomized trials using pure L-theanine supplements concluded that 200 to 400 mg a day may help with stress and anxiety in people under stressful conditions (Williams 2020). A 28-day safety and efficacy trial used 400 mg (Moulin 2024). An anxiety trial in people with generalized anxiety disorder went to 450 to 900 mg (Sarris 2019).

Doses below 200 mg have been tested mainly for brain activity rather than for outcomes you'd notice. The 50 mg alpha study is the clearest example (Nobre 2008). Nothing published tests 100 mg alone against placebo for stress, focus, or sleep in adults.

Calm and the stress response

This is where L-theanine has its most consistent showing, and the strongest results are physiological rather than emotional.

Twelve participants faced a mental arithmetic stressor. L-theanine reduced their heart rate response and their salivary immunoglobulin A response compared with placebo, and heart rate variability analysis pointed to a dampening of sympathetic nervous system activation (Kimura 2007).

Over four weeks, 200 mg daily in 30 healthy adults lowered scores on a depression scale and a trait anxiety scale against placebo, alongside the sleep results below (Hidese 2019). Two of that study's authors work for the company that supplied the tablets.

The systematic review of pure L-theanine trials landed on a cautious version of the same conclusion, that 200 to 400 mg daily may assist stress and anxiety reduction in people exposed to stressful conditions, while calling for longer and larger studies before treating it as established (Williams 2020).

Now the other side. A 28-day trial gave 400 mg daily to 30 adults with moderate stress. Perceived Stress Scale scores fell 17.98 percent on L-theanine and 17.88 percent on placebo, which is effectively the same drop, and salivary cortisol showed no difference between groups (Moulin 2024). In a trial of 46 people with diagnosed generalized anxiety disorder, adjunctive L-theanine at 450 to 900 mg failed to beat placebo on the Hamilton Anxiety scale (p = 0.73) (Sarris 2019). And in 16 volunteers under experimentally induced anxiety, neither 200 mg of L-theanine nor 1 mg of alprazolam produced a measurable effect (Lu 2004).

The pattern that survives all of that: L-theanine turns down physical stress markers in everyday stressful situations. It doesn't treat clinical anxiety, and the subjective calm it produces can be hard to separate from placebo.

Focus, and the caffeine problem

The focus claim is the most commercially useful thing said about L-theanine and the least clean.

Most of the attention research tested L-theanine together with caffeine. A combination of 97 mg L-theanine and 40 mg caffeine improved performance on a task-switching test against placebo, while leaving subjective alertness and intersensory attention unchanged (Einöther 2010). A larger study tested 250 mg L-theanine and 150 mg caffeine separately and together. The combination improved several measures including simple reaction time, working memory reaction time, and sentence verification accuracy (Haskell 2008).

Read that second study carefully, because L-theanine on its own did something different. Alone at 250 mg, it increased headache ratings and reduced the number of correct serial seven subtractions (Haskell 2008). In a separate trial, 200 mg of L-theanine alone slowed overall reaction time on a visual probe task (Rogers 2008).

A study using tea-like ratios, 50 mg L-theanine with 75 mg caffeine, found no effects of L-theanine in isolation at all. Adding it to caffeine cancelled caffeine's vasoconstrictive effect and also eliminated caffeine's improvements to cognition and mood, which the authors noted did not produce a positive result at those levels (Dodd 2015).

So what does L-theanine alone do for cognition? A 2025 meta-analysis of five randomized placebo-controlled trials in 148 healthy adults found a dose-dependent benefit on rapid visual information processing and recognition visual reaction time, a mean difference of 15.20 ms faster. Simple reaction time showed nothing, and the Stroop test showed nothing. The authors titled the paper "Promising, but Not Completely Conclusive" and concluded the benefits could not be confirmed across all test methods (Mátyus 2025). A separate 2025 meta-analysis of 50 randomized trials found L-theanine alone favoured on choice reaction time, with confidence intervals the authors said highlighted uncertainty (Payne 2025). Two of its authors work for tea companies.

The honest summary is that L-theanine alone has a narrow, real effect on some visual attention measures and no reliable effect on others. If you're taking it expecting the focus experience people describe from tea or from theanine-caffeine stacks, the caffeine is doing much of that work.

Sleep

Over four weeks, 200 mg daily lowered Pittsburgh Sleep Quality Index scores against placebo in 30 healthy adults. The subscales that moved were sleep latency, sleep disturbance, and use of sleep medication (Hidese 2019).

In 98 boys aged 8 to 12 with ADHD, 400 mg daily for six weeks raised sleep percentage and sleep efficiency on wrist actigraphy. Sleep latency and the other sleep measures were unchanged (Lyon 2011).

The GAD trial found no benefit on insomnia severity overall (p = 0.35), while participants on L-theanine did report better sleep satisfaction, and a benefit appeared in the subgroup whose insomnia symptoms were below the clinical threshold (Sarris 2019).

A review of the sleep evidence concluded that 200 mg before bed may support sleep quality through anxiolysis rather than sedation, and that L-theanine does not cause daytime drowsiness (Rao 2015). Those authors work for a theanine supplier.

The mechanism matters for expectations. L-theanine appears to work on sleep by lowering the arousal that keeps people awake, which is a different thing from making you sleepy. Detail on timing and dose sits in l-theanine dosage for sleep.

Safety and blood pressure

A review reported the no observed adverse effect level for oral L-theanine above 2,000 mg per kilogram of body weight per day in animal testing (Rao 2015). A 28-day trial at 400 mg daily reported that adverse events resolved and that out of range laboratory values and vital sign changes were judged not clinically relevant (Moulin 2024). Six weeks at 400 mg daily was well tolerated in children (Lyon 2011).

One interaction is worth knowing. In 48 adults, caffeine raised blood pressure and L-theanine antagonised that rise. The authors suggested it may be useful for reducing raised blood pressure (Rogers 2008). Anyone already taking blood pressure medication should raise it with their doctor.

Where the evidence is soft

Sample sizes are small. The four-week adult trial had 30 participants, the stress trial had 30, the GAD trial had 46, and the cognition meta-analysis pooled 148 people across five trials.

Funding is concentrated. Several of the most-cited papers come from companies that make or sell the ingredient, which the article flags where it applies.

And the two 2025 meta-analyses, run by independent and industry-linked teams respectively, both landed on qualified conclusions rather than clear ones (Mátyus 2025, Payne 2025).

What does L-theanine actually do?

It lowers the physical stress response, measurably so on heart rate and salivary markers during a stressor (Kimura 2007). It raises resting alpha brain activity (Nobre 2008). Over weeks it has improved sleep quality and lowered anxiety and depression scale scores in healthy adults (Hidese 2019).

Does it help you focus?

How much do I need?

Reviewers describe the effect as anxiolysis rather than sedation, with no daytime drowsiness (Rao 2015). Resting EEG work points to relaxed alertness (Nobre 2008).

Does it help with anxiety?

For everyday stress, the reviewed evidence is cautiously positive (Williams 2020). For diagnosed generalized anxiety disorder, an 8-week trial at 450 to 900 mg found no benefit over placebo on the primary anxiety measure (Sarris 2019).
